Abstract

This research was conducted at the STABA microbiology laboratory and the Bogor Health Vocational School from January to February 2024. The aim of this research was to determine the comparison of white sweet potato and purple sweet potato media as alternative media for providing nutrients for the growth of S.aureus. with a total of 3 treatments and 6 repetitions with the method using total plate number (ALT). The type of research used is laboratory experimental, and uses the pour plate technique. The growth of S.aureus on white sweet potatoes is round, the size of small co lonies is 0.1cm, the surface is smooth and the color of the colonies is milky white, and the growth of S.aureus on purple sweet potatoes is round, the size of small colonies is 0.1cm, the surface is smooth and the color of the colonies white. This is shown in the growth of S.aureus. It was found that the highest number of white sweet potato bacteria was in the 10% (1.7x106) CFU/ml white sweet potato flour media treatment, while the smallest bacterial growth was in the 5% (4.1x105) CFU/ml purple sweet potato flour media. The results of the research show that white sweet potatoes are one of the best alternative media to use than purple sweet potatoes because the nutritional content and number of bacteria that grow are superior to purple sweet potatoes.

Additional Information: Penelitian ini dilakukan di laboratorium mikrobiologi STABA dan Smk Kesehatan Bogor pada bulan januari sampai februari 2024. Tujuan penelitian ini adalah untuk mengetahui perbandingan media ubi putih dan ubi ungu sebagai media alternatif agar nutrien untuk pertumbuhan S.aureus. dengan jumlah 3 perlakuan dan 6 pengulangan dengan metode yang digunakan angka lempeng total (ALT). Jenis penelitian yang digunakan adalah eksperimental laboratorium, dan menggunakan teknik pour plate. Pertumbuhan S.aureus pada ubi putih berbentuk bulat, ukuran koloni kecil berukuran 0,1cm, permukaan halus dan warna koloni putih susu, dan pertumbuhan S.aureus pada ubi ungu berbentuk bulat, ukuran koloni kecil berukuran 0,1cm, Permukaan halus dan warna koloni putih. Hal ini ditunjukan pada pertumbuhan S.aureus. didapatkan jumlah bakteri ubi putih paling banyak pada perlakuan media tepung ubi putih 10% (1,7x106) CFU/ml sedangkan pertumbuhan bakteri paling kecil pada Media tepung ubi ungu 5% (4,1x105) CFU/ml. Hasil penelitian menunjukan ubi putih termasuk media alternatif yang paling baik digunakan daripada ubi ungu dikarnakan dari kandungan gizi dan jumlah bakteri yang tumbuh lebih unggul dari ubi ungu.
