PERBANDINGAN FIKSASI MENGGUNAKAN NEUTRAL BUFFER FORMALIN 10% DAN PEMANIS ALAMI STEVIA TERHADAP JARINGAN GINJAL TIKUS PUTIH DENGAN PEWARNAAN HEMATOXYLIN EOSIN

Histotechnics is a series of processes for making histological preparations from specimens in the form of animal or human tissue. The stages of the tissue processing process required in making histological preparations are, tissue isolation from body organs, fixation, washing, dehydration, clarification and paraffin infiltration, embedding, cutting, sticking, deparaffinizing, staining. Neutral Buffer Formalin 10% is a solution that is the main standard in anatomical pathology laboratories because the acidity level is close to neutral (pH). The natural sweetener mechanism can be used as an alternative fixative agent to replace Neutral Buffer Formalin 10% because the sucrose content of the aldehyde group will cross-link with amino acids found in the tissue. The aim of this study was to determine the quality of Hematoxylin Eosin staining on rat kidney organs that were fixed using Neutral Buffer Formalin 10% and Stevia natural sweetener. The method used is a static group comparison experiment which aims to determine the changes or effects that arise after certain treatments are carried out. This research used 16 Wistar rat kidneys as test animal subjects. Data obtained from microscopic observations. Data processing used Mann Whithney U. After statistical tests were carried out using the Mann Whithney U test, the research results showed that the results were significant, namely the value of Sig.0.02 < 0.05, which means H1 was accepted and H0 was rejected, so it can be concluded that the quality of fixation in the rat kidney organs that were fixed using the solution Neutral Buffer Formalin 10% better than the results of stevia solution fixation in kidney tissue with Hematoxylin Eosin staining.
